Mouse Retina Analysis

Mouse retina analysis is the systematic examination of retinal structure, cell populations, and molecular features in mice to understand how the visual system forms and functions. Researchers typically collect and section retinal tissue, label specific cell types or gene products with histological or immunofluorescent markers, and use microscopy to quantify spatial organization and developmental changes. In developmental biology, this approach reveals how retinal progenitors differentiate into neurons and supporting cells, how cellular layers are established, and how genetic or environmental conditions alter development. The resulting data support studies of neurogenesis, retinal disease mechanisms, and potential regenerative strategies.

Source: Wang, Z., et al. Transpupillary Two-Photon In Vivo Imaging of the Mouse Retina. J. Vis. Exp. (2021).This video demonstrates a technique for in vivo retinal imaging using two-photon microscopy. Anesthetized mice are prepared for imaging by securing their heads, Applying eye lubricant, and placing a coverslip over the eye. Retinal ganglion cells (RGCs) expressing fluorescent proteins are first visualized using epifluorescence for alignment. Finally, two-photon imaging is employed,...

A method for the incorporation of plasmid DNA into murine retinal cells for the purpose of performing either gain- or loss of function studies in vivo is presented. This method capitalizes on the transient increase in permeability of cell plasma membranes induced by the application of an external electrical field.

The rodent retina has long been recognized as an accessible window to the brain. In this technical paper we provide a protocol that employs the mouse model of oxygen-induced retinopathy to study the mechanisms that lead to failure of vascular regeneration within the central nervous system after ischemic injury. The described system can also be harnessed to explore strategies to promote regrowth of functional blood vessels within the retina and CNS.
