Alkaline Neutralization

Alkaline neutralization is a chemical process that reduces acidity by adding an alkaline substance, helping control pH in industrial and environmental systems. The process occurs when a base, such as sodium hydroxide, calcium hydroxide, or alkaline waste material, reacts with hydrogen ions from an acid to form water and a salt; controlled dosing and mixing bring the solution toward a target pH. In engineering, alkaline neutralization supports wastewater treatment, corrosion control, industrial process management, and remediation of acidic streams. Monitoring pH, alkalinity, reagent demand, and mixing efficiency helps optimize treatment performance while limiting chemical use and secondary waste.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.

An acid-base reaction is one in which a hydrogen ion, H+, is transferred from one chemical species to another. Such reactions are of central importance to numerous natural and technological processes, ranging from the chemical transformations within cells or lakes and oceans to the industrial-scale production of fertilizers, pharmaceuticals, and other substances essential to the society. There are several ways of defining an acid. In the context of aqueous solutions, an acid is a substance...

Cell-Based AAV Neutralizing Antibody Assay: A Colorimetric Technique to Detect Neutralizing Antibodies Against Specific Adeno-Associated Viruses in Serum

2025

In this video, we demonstrate an in vitro colorimetric assay to detect the presence of neutralizing antibodies against alkaline phosphatase-expressing recombinant adeno-associated virus serotype 6 (rAAV6) vectors in sheep serum samples. The assay utilizes the reaction between the AAV encoding a human placental alkaline phosphatase gene and its substrate to generate an insoluble purple product.

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.
