Rhizosphere Analysis

Rhizosphere analysis is the study of the narrow zone of soil influenced by plant roots, where chemical, physical, and biological interactions shape soil and plant health. Researchers examine root exudates, nutrient availability, pH, moisture, microbial communities, and enzyme activity to determine how roots modify their surrounding environment and how soil organisms respond. Sampling methods, microscopy, chemical assays, and molecular tools can reveal nutrient cycling, pollutant transformation, and plant-microbe relationships. In environmental research, rhizosphere analysis helps assess soil fertility, monitor contamination, improve phytoremediation, and predict how vegetation influences ecosystem function under changing conditions.

A Method to Preserve Wetland Roots and Rhizospheres for Elemental Imaging

We describe a protocol to sample, preserve, and section intact roots and the surrounding rhizosphere soil from wetland environments using rice (Oryza sativa L.) as a model species. Once preserved, the sample can be analyzed using elemental imaging techniques, such as synchrotron X-ray fluorescence (XRF) chemical speciation imaging.
