Inflammatory Stimuli

Inflammatory stimuli are biological, chemical, or physical signals that initiate inflammation, a protective response that helps the body detect infection or tissue damage. In immunology, pathogen-associated and damage-associated molecular patterns activate pattern-recognition receptors on immune and tissue cells, triggering signaling pathways such as NF-κB and inflammasome activation, followed by the release of cytokines and chemokines. These mediators recruit leukocytes, alter blood-vessel behavior, and coordinate antimicrobial defense and tissue repair. Studying inflammatory stimuli helps researchers distinguish protective inflammation from harmful chronic responses and supports the development of infection models, biomarkers, and targeted anti-inflammatory therapies.

The distractor-response binding paradigm is described. It can be used to shed light on the influence irrelevant stimuli, competing with targets for a response, can have on human action. Both response retrieval effects and distractor inhibition effects can be analyzed within the paradigm.

Microfluidic flow chambers etched by photolithography and fabricated from PDMS are applied to probe functional outcomes associated with EC dysfunction and inflammation. In a representative experiment, the ability of differential shear stress to modulate monocytic cell adhesion to cytokine activated EC monolayers is demonstrated.

Vascular endothelium tightly controls leukocyte recruitment. Inadequate leukocyte extravasation contributes to human inflammatory diseases. Therefore, searching for novel regulatory elements of endothelial activation is necessary to design improved therapies for inflammatory disorders. Here, we describe a comprehensive methodology to characterize novel endothelial regulators that can modify leukocyte trafficking during inflammation.
