Dss Treatment

DSS treatment is an experimental method that uses dextran sulfate sodium to induce intestinal inflammation, most commonly as a model of colitis in laboratory animals. When administered in drinking water, DSS damages the intestinal epithelial barrier, allowing luminal substances to trigger immune activation, inflammatory signaling, and symptoms such as weight loss, diarrhea, and rectal bleeding. Researchers use this model to study mucosal injury, evaluate disease mechanisms, and assess potential treatments for inflammatory bowel disease. Disease severity can be monitored through clinical signs, colon length, tissue pathology, and inflammatory markers, supporting comparative studies of therapeutic efficacy.

Experimental models of inflammatory bowel disease have allowed us to examine the complex innate and adaptive immune responses associated with pathogenesis. Using histological scoring, quantification of pro-inflammatory cytokines and myeloperoxidase activity, one can begin to assess these responses seen in inflammatory bowel disease.

We demonstrate a protocol in which administration of the genotoxic agent azoxymethane (AOM) followed by three cycles of the pro-inflammatory agent dextran sulfate sodium (DSS) rapidly and consistently generates colon tumors in mice with morphologic and molecular similarities to those seen in human colitis-associated cancer.

Hematoxylin and Eosin Staining to Assess Colon Damage in DSS-Induced Colitis

This video demonstrates the hematoxylin and eosin (H&E) staining method to evaluate colon damage in a murine model of DSS-induced colitis — an inflammatory bowel disease. Upon isolating the colon from a DSS-treated mouse and obtaining sections of the tissue, H&E staining of the sections helps identify the inflammation-mediated tissue damage.

Psoriasis is a chronic, immune-mediated inflammatory skin disease. The Goeckerman regimen, formulated for the treatment of psoriasis, consists of exposure to ultraviolet B (UVB) light and application of crude coal tar (CCT). The following protocol is for the administration of Goeckerman therapy for the treatment of moderate-to-severe psoriasis.
