Dual-probe Detection

Dual-probe detection is a diagnostic strategy that uses two complementary probes to identify the same biological target, improving confidence in measurement and helping distinguish true signals from nonspecific interactions. The probes bind separate sites on a molecule, cell, or tissue, and detection occurs when their combined binding produces a measurable signal, such as fluorescence, luminescence, or another assay readout. In medicine, this approach supports the detection and characterization of biomarkers, pathogens, genetic sequences, and disease-associated cells. By increasing target specificity and enabling signal confirmation, dual-probe methods can strengthen laboratory testing, molecular diagnostics, and research involving disease mechanisms and treatment response.

This protocol describes a general approach to perform photoconversion of fluorescent proteins on a confocal laser scanning microscope. We describe procedures for the photoconversion of puried protein samples, as well as for dual-probe optical highlighting in live cells with mOrange2 and Dronpa.

Early detection of apoptosis may identify at-risk cell populations in a variety of diseases. Here we demonstrate a method to link an early apoptosis-detection protein (Annexin V) to a MRI-detectable iron oxide nanoparticle (SPIO). This method may be extended to other proteins of interest to generate MRI-detectable molecular imaging probes.

Iridium(III) Luminescent Probe for Detection of the Malarial Protein Biomarker Histidine Rich Protein-II

Robust detection reagents are of increasing necessity for developing new malaria diagnostic tools. An iridium(III) probe was designed that emits long-lasting luminescent signal in the presence of a histidine-rich malarial protein biomarker. Detection of the protein either in solution or immobilized on a magnetic particle affords flexibility in application.

Here a method to localize bacteria within paraffin-embedded tissues using DIG-labeled 16S rRNA-targeting DNA probes has been described. This protocol can be applied to study the role of bacteria in various diseases such as periodontitis, cancers, and inflammatory immune diseases.
