Gl261-luc Cells

Gl261-luc cells are a bioluminescent mouse glioma cell line used to model brain tumors and monitor tumor growth in living animals. These cells originate from the GL261 murine glioma model and express luciferase, allowing researchers to visualize tumor burden after administration of a luciferin substrate, which produces detectable light through an enzyme-catalyzed reaction. In medicine, intracranial implantation of Gl261-luc cells supports preclinical studies of glioblastoma biology, tumor progression, immune responses, and therapeutic efficacy. Repeated bioluminescence imaging provides a noninvasive measure of disease development, helping researchers evaluate treatments over time while reducing the need for repeated tissue collection.

In this paper we present a method for transplanting human stem cells into various regions of the central nervous system of the chicken embryo. This provides an in vivo model for assessing the proliferation and differentiation of various types of human stem cells in embryonic tissue environments.

Understanding the malignant behavior of cancer requires creating accurate models of how tumor cells interact with components of the tumor microenvironment, such as macrophages. Here we describe two methods to study glioblastoma cell interaction with tumor associated macrophages and microglia where the effect on glioblastoma invasion is assessed.

Bioluminescence Imaging of an Immunocompetent Animal Model for Glioblastoma

GL261 glioma cells provide a useful immunocompetent animal model of glioblastoma. The goals of this protocol are to demonstrate proper techniques for monitoring intracranial tumor growth using in vivo bioluminescence imaging, and to verify the utility of luciferase-modified GL261 cells for studying tumor immunology and immunotherapeutic approaches for treating glioblastoma.
