Nerve Fiber Density

Nerve fiber density is a quantitative measure of the number or length of nerve fibers within a defined tissue area, providing insight into peripheral nerve structure and function. Researchers estimate it by labeling or visualizing axons in tissue samples, then counting fibers or measuring their total length relative to the examined area using microscopy or image analysis. In medicine, nerve fiber density assessment supports the evaluation of peripheral and small-fiber neuropathies, nerve injury, corneal disease, and treatment responses. Changes in density can help characterize disease progression, compare patient groups, and guide research into nerve repair and regeneration.

In Vivo Single-Fiber Recording of Sciatic Nerve C-Fibers in Rats

This video demonstrates a single-fiber recording of C-fiber nerve conduction in rats. An anesthetized rat is incised to expose the sciatic nerve, which provides sensory and motor supply to the lower limbs. The individual C-fibers are then exposed. Electrical stimulation is provided using stimulus electrodes implanted in the foot, and a response is recorded from a single nerve fiber.

In order to study the changes of nociceptive intraepidermal nerve fibers (IENFs) in painful neuropathies (PN), we developed protocols that could directly examine three-dimensional morphological changes observed in nociceptive IENFs. Three-dimensional analysis of IENFs has the potential to evaluate the morphological changes of IENF in PN.
