Relative Potency Determination

Relative potency determination is a bioassay-based method for comparing the strength of a medicine with that of an established reference preparation. It measures the dose or concentration of each preparation required to produce a defined biological response, then estimates potency from their dose-response relationships under matched assay conditions. Statistical models, including parallel-line or nonlinear curve analysis, can quantify the potency ratio and its uncertainty. In medicine, this approach supports quality control, standardization of biological products, evaluation of drug formulations, and comparison of therapeutic activity when chemical measurements alone do not predict biological effects.

A protocol for the determination of the relative anti-apoptotic activity of an anti-TNFα mAb using a neutralization mechanism with WEHI 164 cells is presented here. This protocol is useful for comparing the neutralization strength of different molecules with the same biological functionality.
