Endotracheal Administration

Endotracheal administration is a method of delivering medication directly into the trachea through an endotracheal tube, providing access to the airway when other routes are unavailable or impractical. After instillation, the substance passes across the tracheal and bronchial lining into the pulmonary circulation, although absorption can be variable and less predictable than intravenous delivery. In medicine, this route has been used in emergency and critical care settings, particularly during resuscitation when vascular access is delayed. Understanding its pharmacokinetic limitations helps clinicians select appropriate routes, doses, and airway-management strategies for time-sensitive treatment.

Non-Invasive Endotracheal Administration of Lipopolysaccharide to Induce Acute Lung Injury in Rodents

2025

In this study, a non-invasive endotracheal instillation technique is demonstrated to develop ALI in rats using a basic laryngoscope to ensure precise LPS delivery to the lungs. The LPS is instilled via an endotracheal tube using a laryngoscope. The magnitude of lung injury is evaluated at multiple time points.

Endotracheal Intubation in Mice via Direct Laryngoscopy Using an Otoscope

We have developed a simple, reliable, and relatively inexpensive method for endotracheal intubation in mice via direct laryngoscopy using an otoscope with a 2.0 mm speculum. This technique is atraumatic and can be used for repeated measurements in chronic experiments. We find it superior to tracheostomy or previously reported nonsurgical techniques.

Self-administration Studies

Behavioral reinforcement induced by the rewarding feelings following substance use sometimes leads to addiction, which is demonstrated by increased self-administration. Drug self-administration studies in rodents model human behavior during drug abuse. These models are useful in understanding the neurobiological behavior of addiction in order to help scientists discover new treatments for drug dependence. This video reviews the concepts underlying self-administration studies. A general...

Many factors need to be evaluated to ensure that the compound is delivered correctly. The route of administration affects the mechanisms of absorption. The characteristics of the substance to be introduced (the pH, viscosity, and concentration) may dictate which route of administration is selected.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN As many research protocols require that a substance be injected into an animal, the route of delivery and the amount of the substance must be accurately determined. There are several routes of administration available in the mouse and rat. Which route to use is determined by several factors of the substance to be injected: the pH of the solution, the volume required for the desired dosage, and...
