Synthetic Mirna Spike

A synthetic miRNA spike is a laboratory-produced microRNA sequence added to a biological sample as an exogenous reference, helping researchers monitor technical variation in miRNA measurements. Introduced at a defined stage of sample processing, it undergoes extraction, reverse transcription, amplification, or sequencing alongside endogenous miRNAs, providing a measurable benchmark for recovery and assay performance. In medicine, synthetic miRNA spikes support more consistent profiling of circulating or tissue-associated miRNAs, strengthen comparisons among samples and experiments, and help distinguish biological changes from workflow-related effects. Their use can improve the reliability of miRNA biomarker studies and the evaluation of diagnostic or therapeutic research methods.

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring.
