Auditory Thalamocortical Slice

An auditory thalamocortical slice is an in vitro brain-tissue preparation that preserves connections between the auditory thalamus and auditory cortex, enabling direct study of communication within this sensory pathway. Researchers prepare a section oriented to retain projections from the medial geniculate body to cortical layers, then use electrophysiology, stimulation, or imaging to measure synaptic transmission and neuronal responses under controlled conditions. This model helps characterize thalamocortical circuit organization, sensory information processing, and activity-dependent plasticity while reducing the complexity of the intact brain. Findings can clarify mechanisms underlying auditory perception and provide a foundation for investigating circuit changes associated with neurological disorders.

Preparation of Colliculo-Thalamocortical Slices from a Mouse Pup Brain

2025

The video demonstrates the preparation of colliculo-thalamocortical brain slices from mouse pup brains. An isolated brain is aligned on a marked slide and cut at an angle to align the auditory midbrain and forebrain in the same plane. The brain is then sliced using a vibratome, and the slices containing the major auditory midbrain and forebrain structures are transferred to a holding chamber for preservation.

The chicken auditory brainstem is comprised of nuclei responsible for binaural sound processing. A single coronal slice preparation maintains the entire circuitry while the cultured approach provides a unique preparation to study the development of neuronal structure and auditory function at the molecular, cellular and network levels.

Preparing Wedge Brain Slices to Visualize Auditory Neurons

This video demonstrates the process of preparing wedge-shaped brain slices in a laboratory setting. Using a vibratome, a wedge-shaped slice from a mouse's brain is crafted with varying thickness. This slice contains the auditory nerve root positioned on the thicker end, while the hearing-related neurons are located on the thinner end.
