Brain Targeting

Brain targeting is the deliberate delivery of drugs, genes, imaging agents, or other therapeutic materials to specific regions or cell types in the brain. Because the blood-brain barrier restricts entry from the circulation, targeting strategies use approaches such as receptor-mediated transport, engineered nanoparticles, intranasal administration, or localized barrier opening to improve delivery and reduce exposure elsewhere. In neuroscience, these methods support studies of neural circuits, disease mechanisms, and treatment response while addressing conditions including tumors, neurodegenerative disorders, and brain injury. Improved targeting can increase therapeutic precision, limit systemic side effects, and advance personalized brain medicine.

Controlling and analyzing neural circuits in vivo would be facilitated by a technology for delivery of viruses and other reagents to desired 3-dimensional sets of brain regions. We demonstrate customized fluidic injector array fabrication, and delivery of virally-encoded optical sensitizers, enabling optical manipulation of complex brain circuits.

Source: Zhang, S., et al. Intracranial Subarachnoidal Route of Infection for Investigating Roles of Streptococcus suis Biofilms in Meningitis in a Mouse Infection Model. J. Vis. Exp. (2018).This video demonstrates the detection of target gene expression in a bacteria-infected mouse brain. RNA is extracted, treated with a DNA-degrading enzyme, and reverse-transcribed into cDNA. The cDNA is amplified through thermal cycling with gene-specific primers, while a DNA-binding dye binds to the...

2025

In this video, we demonstrate the electrochemiluminescence immunoassay for the quantitation of target protein levels in different samples. This assay is based on the luminescence produced in the electrochemical reaction. The intensity of light emitted is proportional to the quantity of the target protein in the sample.
