Cortical Vasculature Imaging

Cortical vasculature imaging is the visualization and measurement of blood vessels supplying the cerebral cortex, a key approach for studying how vascular structure supports neural function. Imaging methods such as two-photon microscopy, fluorescence-based angiography, and magnetic resonance imaging reveal vessel architecture, blood flow, and changes in oxygenation by detecting labeled blood, endogenous signals, or hemodynamic contrasts. In neuroscience, these measurements help link neuronal activity to neurovascular responses and characterize processes such as vessel remodeling, blood-brain barrier disruption, ischemia, and tumor growth. The resulting maps and flow data support research into brain physiology, disease mechanisms, and potential therapeutic strategies.

In vivo Imaging of Deep Cortical Layers using a Microprism

Right-angle microprisms inserted into the mouse neocortex allows for deep imaging of multiple cortical layers with a viewpoint typically found in slice. One-millimeter microprisms offer a wide field-of-view (~900 μm) and spatial resolutions sufficient to resolve dendritic spines. We demonstrate layer V neuronal imaging and neocortical vascular imaging using microprisms.

Calcium signals play a key role in many cellular processes including gene expression, survival and differentiation. Here we demonstrate how to perform calcium imaging using Fura-2 AM. Calcium imaging is a valuable tool to study the regulation of intracellular calcium in real time and its regulation of signaling cascades.

A high resolution ex vivo 7T MR imaging protocol is presented, to perform MR-guided histopathological validation of microvascular pathology in post-mortem human brain tissue. Furthermore, guidelines are provided for the assessment of cortical microinfarcts on in vivo 7T as well as 3T MR images.

Measuring gyrification (cortical folding) at any age represents a window into early brain development. Hence, we previously developed an algorithm to measure local gyrification at thousands of points over the hemisphere1. In this paper, we detail the computation of this local gyrification index.
