Fm Dye Imaging

Fm dye imaging is a fluorescence-based technique for visualizing synaptic vesicle recycling, making it valuable for studying how neurons release and retrieve neurotransmitter-filled vesicles. Lipophilic Fm dyes insert into the plasma membrane during vesicle endocytosis, label the vesicle membrane, and lose fluorescence when vesicles fuse with the cell surface and release the dye into the surrounding solution. In neuroscience, this approach measures presynaptic exocytosis, endocytosis, and recycling dynamics in cultured neurons and neuromuscular preparations. By linking fluorescence changes to synaptic activity, Fm dye imaging helps reveal how stimulation, calcium signaling, and pharmacological interventions regulate neurotransmission.

FM Dyes in Vesicle Recycling

2023

FM dyes are a class of fluorescent molecules that has found important use in studying the vesicle recycling process. By virtue of a chemical structure, these molecules can insert themselves into the outer leaflet of phospholipid bilayer membranes. After membrane insertion, they are internalized into the cell via endocytosed vesicles, and released when these vesicles recycle back to the membrane. Since, these dyes fluoresce strongly in the hydrophobic environment within membranes and weakly in...

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.

Electronic pre-resonance stimulated Raman scattering (epr-SRS) imaging of rainbow-like Raman dyes is a new platform for highly multiplexed epitope-based protein imaging. Here, we present a practical guide including antibody preparation, tissue sample staining, SRS microscope assembly, and epr-SRS tissue imaging.
