Genetically Modified Mouse Pup

A genetically modified mouse pup is a newborn mouse carrying an experimentally introduced, deleted, or altered genetic sequence, providing a living model for studying gene function. Researchers generate these pups by modifying embryos, often with CRISPR-Cas9 or transgene insertion, so the resulting DNA change is incorporated into developing cells and may be inherited by offspring. In neuroscience, genetically modified mouse pups help link specific genes to neuronal development, synaptic signaling, behavior, and neurological disease by enabling analysis across molecular, cellular, and whole-animal levels. Their phenotypes can guide studies of disease mechanisms and potential treatments, while genotyping and appropriate controls help confirm that observed effects arise from the intended modification.

2023

Source: Laboratories of Margaret Workman and Kimberly Frye - Depaul University Genetic modification of foods has been a controversial issue due to debated concerns over health and environmental safety. This experiment demonstrates technical understanding of how food DNA is genetically identified, allowing for educated decision making about the safety and potential dangers of using genetically modified organisms (GMOs) in food supplies. Polymerase Chain Reaction (PCR) is used to amplify food DNA...

The microinjection of mouse oocytes is commonly used for both classic transgenesis (i.e., the random integration of transgenes) and CRISPR-mediated gene targeting. This protocol reviews the latest developments in microinjection, with a particular emphasis on quality control and genotyping strategies.

Source: Quattrocolo, G., et al. Homochronic Transplantation of Interneuron Precursors into Early Postnatal Mouse Brains. J. Vis. Exp. (2018)This video demonstrates the transplantation of interneuron precursor cells into the neonatal mouse brain using either a homotopic or heterotopic approach. The procedure involves securing an anesthetized recipient pup, identifying the target brain region, and injecting a suspension of precursor cells derived from a donor pup at the same developmental stage.
