Malachite Green Reagent

Malachite Green Reagent is a colorimetric chemical reagent used in biochemistry to detect and quantify inorganic phosphate, making it valuable for measuring phosphate-releasing reactions. In acidic conditions, phosphate reacts with molybdate to form a phosphomolybdate complex, which associates with malachite green and produces an intensely green color; absorbance increases with phosphate concentration. By comparing sample absorbance with standards, researchers can determine phosphate levels or monitor enzyme activities such as phosphatases and ATPases. This sensitive assay supports biochemical studies of enzyme kinetics, substrate turnover, and cellular phosphate metabolism without requiring radioactive labeling.
