Bead-based Proximity Assay

A bead-based proximity assay is a biochemical method that detects molecular interactions or analytes by using functionalized beads to bring reaction partners into close physical proximity. Beads coated with antibodies, nucleic acids, or other binding molecules capture target components, and their proximity enables a measurable signal through processes such as enzymatic reaction, fluorescence, or nucleic acid amplification. These assays support sensitive detection of proteins, nucleic acids, and biomolecular complexes in solution, often with multiplexing and straightforward separation of bead-bound material. They are valuable for studying molecular associations, quantifying low-abundance targets, and developing diagnostic and high-throughput research workflows.

Protein-protein interactions can occur in both the nucleus and the cytoplasm of a cell. To investigate these interactions, traditional co-immunoprecipitation and modern proximity ligation assay are applied. In this study, we compare these two methods to visualize the distribution of NF90-RBM3 interactions in the nucleus and the cytoplasm.

Detection of Protein-Protein Interactions in Drosophila Larvae Using a Proximity Ligation Assay

This video demonstrates the visualization of protein-protein interactions at the Drosophila neuromuscular junction (NMJ) using Proximity Ligation Assay (PLA). After isolating the body walls, interacting proteins are probed with primary antibodies and PLA-conjugated secondary antibodies. The proximity of the interacting proteins facilitates the ligation of probes with connector oligonucleotides, forming circular DNA, which is then amplified by rolling circle amplification. Fluorescent probes...
