Blood Sample Separation

Blood sample separation is the process of isolating blood components, such as plasma, serum, red blood cells, white blood cells, and platelets, for analysis or further use. In conventional workflows, centrifugation separates components according to density, causing cells to form distinct layers while less dense plasma or serum remains above them; filtration and microfluidic methods can provide additional size- or flow-based control. In bioengineering, these approaches support diagnostic testing, biomarker measurement, cell-based research, transfusion preparation, and development of blood-processing devices. Reliable separation improves sample quality, reduces cross-contamination, and helps researchers obtain reproducible biological measurements.

We describe a method combining immunomagnetic beads and fluorescence-activated cell sorting to isolate and analyze defined immune cell subpopulations of peripheral blood mononuclear cells (monocytes, CD4+ T cells, CD8+ T cells, B cells, and natural killer cells). Using this method, magnetic and fluorescently labeled cells can be purified and analyzed.
