Droplet Generation Device

A droplet generation device is an instrument that produces discrete, uniform liquid droplets from a continuous phase, enabling precise control of small-volume reactions and materials. In bioengineering, it commonly uses microfluidic channels to bring immiscible fluids together, while interfacial forces, flow rates, and channel geometry cause one phase to break into droplets within another. Adjusting these parameters controls droplet size, spacing, and production rate, while allowing cells, biomolecules, or reagents to be encapsulated in isolated compartments. These capabilities support single-cell analysis, digital assays, drug screening, biomaterial synthesis, and other applications requiring reproducible microscale environments.

This microfluidic system has the potential to be used in a wide variety of biological applications given its low cost, ease of use, volumetric precision, high speed, repeatability and automation.

Microfluidic double emulsions generation typically involves devices with patterned wettability or custom-fabricated glass components. Here we describe the fabrication and testing of an all polydimethylsiloxane (PDMS) double emulsion generator that does not require surface treatment or complicated fabrication processes, and is capable of producing double emulsions down to 14 µm.

We describe a protocol for the microfabrication of the gradient-generating microfluidic device that can generate spatial and temporal gradients in well-defined microenvironment. In this approach, the gradient-generating microfluidic device can be used to study directed cell migration, embryogenesis, wound healing, and cancer metastasis.

The video demonstrates the formation of neuromuscular junctions using a microfluidic device. Neural precursor cells and muscle progenitor cells are seeded into different wells within the device and left to mature into motor neurons and myotubes, respectively.
