Metabolic Perturbation

Metabolic perturbation is the deliberate alteration of an organism’s or cell’s metabolic state to examine how biochemical pathways respond and function. In bioengineering, researchers change nutrient availability, enzyme activity, gene expression, or pathway inputs, then measure shifts in metabolite levels, flux, energy use, or cellular behavior. These interventions help reveal pathway regulation, identify metabolic bottlenecks, and connect genotype with phenotype. Metabolic perturbation supports the design of engineered microbes and cells for producing valuable compounds, optimizing bioprocesses, studying disease-associated metabolism, and evaluating how biological systems adapt to controlled changes.

Metabolic Profile Analysis of Zebrafish Embryos

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

Overview Metabolism represents all of the chemical activity in a cell, including reactions that build molecules (anabolism) and those that break molecules down (catabolism). Anabolic reactions require energy, whereas catabolic reactions provide it. Optogenetic Perturbation of Neural Activity with Laser Illumination in Semi-intact Drosophila Larvae in Motion

Here we describe a protocol for optogenetic manipulation of motoneuronal activity while monitoring changes in motor output (muscle contraction) in semi-intact Drosophila larvae using lasers within a conventional confocal microscope. This technique enables researchers to achieve local perturbation of neural activity in a few neuromeres to elucidate the dynamics of motor circuits.
