Perfusion Devices

Perfusion devices are systems that circulate nutrient- and oxygen-containing fluids through organs, tissues, or engineered constructs to maintain viable, controlled physiological conditions. They use pumps, tubing, reservoirs, and monitoring components to regulate fluid flow, pressure, temperature, and composition, enabling mass transport that diffusion alone cannot provide in larger samples. In bioengineering, these devices support ex vivo organ preservation, vascularized tissue culture, bioreactor development, and studies of drug delivery or tissue function. By reproducing aspects of blood circulation, perfusion systems improve experimental control and help researchers evaluate engineered tissues, disease models, and potential therapeutic interventions.

The protocol describes how to engineer a perfusable vascular network in a spheroid. The spheroid's surrounding microenvironment is devised to induce angiogenesis and connect the spheroid to the microchannels in a microfluidic device. The method allows the perfusion of the spheroid, which is a long-awaited technique in three-dimensional cultures.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

2025

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.

Here we present a protocol describing oxygenated ex situ machine perfusion of donor liver grafts. This article contains a step by step protocol to procure and prepare the liver graft for machine perfusion, prepare the perfusion fluid, prime the perfusion machine and perform oxygenated normothermic machine perfusion of the liver graft.
