Photolithography Lift-off

Photolithography lift-off is a microfabrication technique that creates patterned thin films by depositing material onto a prepatterned sacrificial photoresist and then removing the resist. During processing, light defines features in the resist through a mask, development exposes selected substrate regions, and metal or another functional material coats both the substrate and resist; dissolving the resist removes the overlying material while leaving the desired pattern behind. In bioengineering, this approach produces electrodes, microfluidic features, biosensor surfaces, and cell-guiding patterns with controlled dimensions, supporting lab-on-a-chip devices, neural interfaces, and other microscale systems where direct etching could damage underlying materials.

This widely used method utilizes light to transfer a pattern onto a silicon wafer, and provides the basis for the fabrication of many types of BioMEM devices. Multilayer microfluidic devices often involve the fabrication of master molds with complex geometries for functionality. This article presents a complete protocol for multi-step photolithography with valves and variable height features tunable to any application. As a demonstration, we fabricate a microfluidic droplet generator capable of producing hydrogel beads.
