Transverse Axial Tubule System

The transverse axial tubule system is a network of sarcolemmal invaginations in striated muscle cells that carries electrical signals deep into the cell, helping coordinate contraction. In cardiac myocytes, transverse and axial tubules position L-type calcium channels near ryanodine receptors on the sarcoplasmic reticulum, enabling action potential-triggered calcium-induced calcium release and synchronized excitation-contraction coupling. In bioengineering, analyzing this nanoscale architecture supports the design and evaluation of engineered heart tissues, computational models, and therapeutic strategies for restoring cellular electrical organization and contractile function. Changes in tubule structure can also provide indicators of cardiac dysfunction and tissue maturation.

This video demonstrates the isolation procedure of Sertoli cells from mouse seminiferous tubules, a component of the testes. Enzymatic digestion and repeated washes promote the release of Sertoli cells into the solution, which can be cultured further for proliferation and growth.

Method to Measure Tone of Axial and Proximal Muscle

We have developed a device (Twister) to study the regulation of tonic muscle activity during active postural maintenance. Twister measures torsional resistance and muscular responses in standing subjects during twisting of the body axis. The device can be flexibly configured to study various aspects of tonic control across the neck, trunk, and/or hips.
