Actin Cortex Reconstitution

Actin cortex reconstitution is an in vitro method for rebuilding the thin, contractile actin network that supports cell membranes, allowing its organization and mechanics to be studied under controlled conditions. In a typical system, purified actin polymerizes at a membrane or membrane mimic through anchoring proteins, while cross-linkers and myosin motors tune network architecture, force generation, and contractility. These minimal models help separate the contributions of actin assembly, membrane attachment, and motor activity to cell shape, polarity, adhesion, migration, and cytokinesis. The approach also supports synthetic-cell design and quantitative tests of how cytoskeletal forces produce cellular behavior.

Filamentous actin (F-actin) plays an important role in spinogenesis, synaptic plasticity, and synaptic stability. Quantification of F-actin puncta is therefore a useful tool to study the integrity of synaptic structures. This protocol describes the procedures of quantifying F-actin puncta labeled with Phalloidin in low-density primary cortical neuronal cultures.
