Ammonium Acetate Elution

Ammonium acetate elution is a laboratory technique that uses an ammonium acetate solution to release charged molecules from a separation matrix, supporting the purification of biological samples. Its mechanism depends on increased ionic strength: ammonium and acetate ions compete with nucleic acids, proteins, or other analytes for charged binding sites, weakening electrostatic interactions and promoting elution. Because ammonium acetate is relatively volatile, it can often be removed readily after collection, helping preserve purified material for downstream analysis. In biology, the method is useful in nucleic acid purification, chromatography, and sample preparation for biochemical and mass spectrometric workflows.

Direct Detection of the Acetate-forming Activity of the Enzyme Acetate Kinase

A method for the determination of acetate kinase activity is described. This assay utilizes a direct reaction for determining enzyme activity and kinetics of acetate kinase in the acetate-forming direction with different phosphoryl acceptors. Furthermore, this method can be utilized for assaying other acetyl phosphate or acetyl-CoA utilizing enzymes.

We demonstrate the extraction of ammonium from an ammonium-rich stream using an electrochemical and a bioelectrochemical system. The reactor setup, operation and data analysis are discussed.
