Antegrade Perfusion

Antegrade perfusion is a technique that delivers blood, culture medium, or preservation solution through an organ’s arterial supply in the direction of normal physiological flow. A pump or controlled pressure gradient drives the perfusate through arteries into arterioles and capillaries, allowing oxygen, nutrients, or protective compounds to reach the tissue while metabolic waste is removed. In biology and biomedical research, antegrade perfusion supports isolated-organ studies, tissue preservation, transplantation workflows, and investigations of vascular function. By maintaining more natural circulation patterns, it can improve tissue viability and provide a controlled system for assessing organ responses outside the body.

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.

We developed a simple method for isolating high quality individual mouse heart cells by the antegrade perfusion technique. This method is Langendorff-free and useful for isolating ventricular and atrial myocytes or interstitial cells, such as cardiac fibroblasts or progenitors.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair. It holds a high translational value, however it is technically challenging and labor-intensive.
