Cytokinesis

Cytokinesis is the final stage of cell division, when a parent cell’s cytoplasm separates to form two daughter cells, making it essential for growth, development, and tissue maintenance. In animal cells, an actin-myosin contractile ring tightens beneath the plasma membrane to create a cleavage furrow, whereas plant cells assemble vesicles into a cell plate that expands outward and becomes a new cell wall. Coordinated with chromosome segregation and nuclear division, cytokinesis preserves cellular organization and genome distribution; defects can produce abnormal cell numbers, multinucleation, or aneuploidy, making the process important in developmental biology and disease research.

In eukaryotes, the cell division cycle is divided into distinct, coordinated cellular processes that include cell growth, DNA replication/chromosome duplication, chromosome distribution to daughter cells, and finally, cell division. The cell cycle is tightly regulated by its regulatory systems as well as extracellular signals that affect cell proliferation. The processes of the cell cycle occur over approximately 24 hours (in typical human cells) and in two major distinguishable stages. The...

This protocol describes how to image dividing cells within a tissue in Caenorhabditis elegans embryos. While several protocols describe how to image cell division in the early embryo, this protocol describes how to image cell division within a developing tissue during mid late embryogenesis.
