Embryonic Kidney

The embryonic kidney is the developing organ that establishes the structures and functions required for filtration, fluid balance, and waste removal after birth. It forms from intermediate mesoderm through reciprocal signaling between the ureteric bud and metanephric mesenchyme: the bud branches to create the collecting duct system, while induced mesenchymal cells differentiate into nephrons. This coordinated process involves cell differentiation, tissue patterning, and vascular development, and disruptions can cause congenital kidney abnormalities. Studying embryonic kidney development helps explain renal disease, guides research on kidney organoids and regenerative medicine, and provides models for investigating how organs form.

In this report we describe a method for the isolation and culture of the progenitor cell niche from the embryonic mouse kidney that can be used to study signaling pathways regulating stem/progenitor cells of the developing kidney. These cultured cells are highly accessible to small molecule and recombinant protein treatment, and importantly also to viral transduction, which allows efficient manipulation of candidate pathways.

Source: Lucas-Hourani, M. et.al., High-throughput Screening for Broad-spectrum Chemical Inhibitors of RNA Viruses. J. Vis. Exp. (2014)This video shows a luciferase-based antiviral assay in human embryonic kidney cells infected with recombinant measles virus. It shows that serially diluted test compounds inhibit viral replication, leading to lower luminescence in a dose-dependent manner while maintaining host cell viability.
