Gel Mold Assembly

Gel mold assembly is the process of fitting mold components together to create a defined cavity that contains a liquid gel as it forms, enabling reproducible shapes and dimensions in biological research. The assembled mold controls the gel’s boundaries during physical or chemical gelation, while alignment and sealing help prevent leakage, deformation, and unwanted variation between samples. In biology, researchers use gel molds to prepare structured hydrogels, tissue-engineering constructs, cell-laden matrices, and experimental substrates. Consistent assembly supports reliable sample handling, imaging, mechanical testing, and comparisons across experiments, making it an important practical step in studies of biomaterials, cell behavior, and three-dimensional culture.

We have devised a method for low-cost and rapid prototyping of liquid elastomer rubber injection molded devices by using fused deposition modeling 3D printers for mold design and a modified desiccator as a liquid injection system.

Fabrication of Custom Agarose Wells for Cell Seeding and Tissue Ring Self-assembly Using 3D-Printed Molds

This protocol describes a platform for fabricating self-assembled tissue rings in variable sizes using a customized 3D-printed plastic mold. PDMS negatives are cured in the 3D-printed mold; then agarose is cast in the cured PDMS negatives. Cells are seeded into the resulting agarose wells where they aggregate into tissue rings.

This manuscript presents an injection molding method to engineer microvessels that recapitulate physiological properties of endothelium. The microfluidic-based process creates patent 3D vascular networks with tailorable conditions, such as flow, cellular composition, geometry, and biochemical gradients.
