Gelatinase Detection

Gelatinase detection is the identification or measurement of enzymes that hydrolyze gelatin, a denatured form of collagen, and is important for studying extracellular protein degradation in biology. In a typical assay, gelatin serves as the substrate; active gelatinases cleave its peptide bonds, producing clear zones in gelatin-containing media or bands of reduced staining in substrate gels after electrophoresis. These methods help distinguish gelatinolytic activity, estimate enzyme abundance, and compare activity under different conditions. Gelatinase detection supports research on microbial virulence, extracellular matrix remodeling, wound healing, inflammation, and cancer biology, where altered protease activity can affect tissue structure and disease progression.

2025

Proteinases are vital for bacterial pathogenesis, enabling invasion and immune evasion. Single-step extraction of cytoplasmic and membrane proteins of bacteria in their active form is harrowing. This video article demonstrates the extraction of whole cellular proteins of Leptospira for zymography analysis to detect and characterize gelatinases.

2025

This video demonstrates the zymography gel electrophoresis technique to assess the activity of matrix metalloproteinases (MMPs) present in cell culture supernatant. The active MMPs degrade the MMP-specific substrate embedded in a polyacrylamide gel, which appears as clear bands against a dark stained background following staining.

Ubiquitination is a key posttranslational modification carried out by a set of three enzymes. Mutations of genes involved in this modification are associated with many different human diseases. Here, we describe protocols to detect protein ubiquitination in cultured cells in vivo and test tubes in vitro.
