Gonad Extrusion Technique

The gonad extrusion technique is a biological dissection method that releases gonadal tissue from an intact organism for direct laboratory examination. After the specimen is immobilized, a controlled incision or opening allows gentle pressure to extrude the gonad while minimizing damage to its cellular organization. The exposed tissue can then be mounted for microscopy, stained to identify cell types or developmental stages, or analyzed for changes in morphology and gene expression. By providing access to reproductive tissue without relying solely on whole-organism imaging, this technique supports studies of gonad development, gametogenesis, reproductive biology, and the cellular effects of genetic or environmental conditions.

Dying cells are extruded from epithelial tissues by concerted contraction of neighboring cells without disrupting barrier function. The optical clarity of developing zebrafish provides an excellent system to visualize extrusion in living epithelia. Here we describe methods to induce and image extrusion in the larval zebrafish epidermis at cellular resolution.
