Human T-cell Assays

Human T-cell assays are laboratory methods that measure how human T lymphocytes recognize stimuli and carry out immune functions, making them important for studying adaptive immunity, infection, cancer, and therapeutic responses. These assays typically isolate or culture T cells, expose them to antigens or activating signals, and quantify outcomes such as proliferation, cytokine release, cytotoxicity, or surface-marker expression through methods including flow cytometry and immunoassays. Researchers use these measurements to characterize antigen-specific responses, evaluate vaccines and immunotherapies, investigate immune disorders, and compare cellular activity across conditions. Careful control of donor samples, culture conditions, and stimulation protocols improves reproducibility and interpretation.

Luciferase Assay for Measuring γ-Secretase Activity in Human Cells

2025

This video demonstrates the procedure of measuring γ-secretase activity in genetically modified human cells using a tetracycline-inducible system. The process involves inducing APP gene expression with tetracycline, generating amyloid-β peptides, and activating luciferase production. The bioluminescence resulting from the luciferase-substrate reaction is then measured, reflecting γ-secretase activity.

Colony Forming Cell (CFC) Assay for Human Hematopoietic Cells

The colony forming cell (CFC) assay is an in vitro assay in which hematopoietic progenitors form colonies in a semi-solid medium. A combination of colony morphology, cell morphology, and flow cytometry are used to assess the ability of the progenitors to proliferate and differentiate along the different hematopoietic lineages.

In this video, we demonstrate an alternative method for detection and titering of viruses using an enzymatic antigen detection technique known as an immunoperoxidase assay. Here, we will show you how to collect your viral samples, prepare the cells for testing, and finally the immunoperoxidase assay using serial dilutions to determine the viral titer.
