Low Phototoxicity

Low phototoxicity refers to minimizing biological damage caused by light exposure during imaging or other light-based experiments. In fluorescence microscopy, it is achieved by reducing excitation intensity and exposure time, selecting suitable wavelengths and fluorophores, and limiting repeated illumination, which helps decrease the formation of reactive oxygen species, cellular stress, and photobleaching. These strategies preserve cell viability and normal behavior while maintaining useful image quality, making low-phototoxicity approaches important for live-cell imaging, developmental biology, long-term time-lapse studies, and quantitative measurements of dynamic cellular processes.

Photodynamic therapy (PDT) is a non-invasive and non-surgical method for lung cancer treatment. Photosensitizers selectively accumulate in tumor tissue and lead to tumor cell death in the presence of oxygen and the proper wavelength of light. This protocol describes a rod-based approach, combining 3D-printing and soft lithography techniques for fabricating the soft gripper devices. This approach eliminates the need for an external air source by incorporating a chamber component and reduces the chance of occlusion during the sealing process, particularly for miniaturized pneumatic channels.

The cholesterol assay is designed to quantitate the rate of cholesterol efflux from cultured cells and the capacity of plasma acceptors to accept cholesterol released from cells. The assay consists of labelling cells with cholesterol, equilibration of cholesterol among intracellular pools and release of cholesterol to an extracellular acceptor.
