Lymphoid Organ Population

Lymphoid organ population refers to the collection and distribution of immune cells within lymphoid organs, including the bone marrow, thymus, lymph nodes, spleen, and mucosa-associated tissues. These populations arise through lymphocyte development, maturation, migration, and antigen-driven expansion: precursor cells develop in primary lymphoid organs, while secondary organs organize encounters among lymphocytes, antigen-presenting cells, and foreign molecules. Characterizing lymphoid organ populations helps researchers understand immune system development, pathogen defense, inflammation, and immune disorders. Population analysis using microscopy, flow cytometry, or molecular profiling can reveal changes in cell composition and organization during infection, vaccination, aging, or disease.

This video demonstrates an imaging method to visualize in situ interferon-gamma (IFN-γ) secretion in the mouse spleen. Following bacterial infection and the induction of IFN-γ accumulation inside T cells and natural killer cells, the spleen is harvested from the mouse. Upon obtaining tissue sections, immunostaining is performed to visualize IFN-γ inside the cells.
