Mature Mirna

Mature miRNA is a short, processed noncoding RNA molecule that regulates gene expression after transcription, making it an important mediator of cellular development, physiology, and disease. In animals, RNA polymerase II first produces a primary miRNA transcript; Drosha and DGCR8 cleave it into a precursor hairpin, Exportin-5 transports the precursor to the cytoplasm, and Dicer generates a short RNA duplex whose guide strand loads into Argonaute-containing RISC. The complex recognizes partially complementary sequences, often in target mRNA 3′ untranslated regions, to reduce translation or promote mRNA destabilization. Studying mature miRNA supports biomarker development, functional genetics, and therapeutic research.

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring. This method will help identify new circulating biomarkers capable of predicting clinical outcomes in patients.
