Oocyte Developmental Competence

Oocyte developmental competence is the ability of an oocyte to mature, undergo successful fertilization, and support development into a viable embryo. This capacity develops through coordinated nuclear and cytoplasmic maturation, including chromosome organization, organelle function, energy production, and accumulation of maternal RNAs and proteins that guide early embryogenesis before embryonic genome activation. Assessing developmental competence helps researchers understand fertility, reproductive aging, and causes of early developmental failure. In assisted reproductive technologies, competence-related markers can inform oocyte selection, culture strategies, and embryo development outcomes, while studies of oocyte biology support advances in infertility treatment, cryopreservation, and reproductive biotechnology.

Here, we present a protocol for non-invasive assessment of oocyte developmental competence performed during their in vitro maturation from the germinal vesicle to the metaphase II stage. This method combines time-lapse imaging with particle image velocimetry (PIV) and neural network analyses.

Functional Cloning Using a Xenopus Oocyte Expression System

We describe a Xenopus oocyte and animal cap system for the expression cloning of genes capable of inducing a response in competent ectoderm, and discuss techniques for the subsequent analysis of such genes. This system is useful in the functional identification of a wide range of gene products.
