Stromal Fibroblasts

Stromal fibroblasts are connective-tissue cells that produce and organize the extracellular matrix, helping maintain tissue structure and regulate the local cellular environment. They synthesize components such as collagen and fibronectin and communicate with nearby cells through secreted signaling molecules, allowing them to influence inflammation, repair, and tissue remodeling. In biology and biomedical research, stromal fibroblasts are studied in wound healing, fibrosis, immune regulation, and tumor development, where changes in their activity can alter tissue stiffness and cell behavior. Understanding these cells supports research into tissue regeneration, disease mechanisms, and therapies that target abnormal tissue remodeling.

Experimental Generation of Carcinoma-Associated Fibroblasts (CAFs) from Human Mammary Fibroblasts

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

In this report, we demonstrate a system to isolate and culture donor cells from the mouse mammary gland, and orthotopically transplant these cells in recipient mice to analyze stromal: epithelial interactions during mammary tumor development.

Here, a protocol to isolate and establish primary fibroblast/myofibroblast (MF) cultures from frozen gastric, small intestinal, and colonic tissue-yielding cells with a MF phenotype-is presented. These cells express CD90, α-SMA and vimentin. MFs can be used for a variety of functional assays including enzymatic activity and cytokine production.

We describe methodologies for establishing in vitro heterotypic three-dimensional models comprising ovarian fibroblasts and normal ovarian surface or ovarian cancer epithelial cells. We discuss the use of these models to study stromal-epithelial interactions that occur during ovarian cancer development.
