Foxo1 Localization Study

A FoxO1 localization study examines where the transcription factor FoxO1 resides within a cell and how its distribution relates to cancer biology. FoxO1 shuttles between the nucleus and cytoplasm in response to signaling, particularly through Akt-mediated phosphorylation, which can promote its export from the nucleus and alter transcription of genes involved in cell-cycle control, apoptosis, metabolism, and stress responses. Researchers assess localization using methods such as immunofluorescence microscopy, cell fractionation, or tagged-protein imaging. These studies help clarify how disrupted FoxO1 trafficking influences tumor development, treatment response, and the identification of potential therapeutic targets.

Using an α-Bungarotoxin Binding Site Tag to Study GABA A Receptor Membrane Localization and Trafficking

Here we demonstrate the use of fluorescent Alexa dye coupled to α-bungarotoxin to measure GABA A receptor surface localization and endocytosis in hippocampal neurons. Through the use of constructs bearing a short extracellular tag that binds α-bungarotoxin, analysis of plasma membrane protein endocytic trafficking can be achieved.

An In Vivo Immunofluorescence Localization Method to Study Antibody Biodistribution

This video demonstrates an in vivo immunofluorescence localization (IVIL) technique to study the biodistribution of tumor-targeting antibodies in a murine model of breast cancer. Upon the intravenous injection of a tumor-specific antibody in the mouse, the tumor is surgically isolated, and the localization of the antibody within the tumor tissue is confirmed via ex vivo immunostaining.

Isolation of mRNAs Associated with Yeast Mitochondria to Study Mechanisms of Localized Translation

Many mRNAs encoding mitochondrial proteins are associated with the mitochondria outer membrane. We describe a subcellular fractionation procedure aimed at isolation of yeast mitochondria with its associated mRNAs and ribosomes. This protocol can be applied to cells grown under diverse conditions in order to reveal mechanisms of mRNA localization and localized translation near the mitochondria.

Local Application of Drugs to Study Nicotinic Acetylcholine Receptor Function in Mouse Brain Slices

In this paper, we describe a useful method to study ligand-gated ion channel function in neurons of acutely isolated brain slices. This method involves the use of a drug-filled micropipette for local application of drugs to neurons recorded using standard patch clamp techniques.

Here, we outline how to study mitochondrial localization of a (cell cycle) kinase, and how to determine its sub-mitochondrial location as well as potential mitochondrial substrates/targets. Forced expression of proteins into the mitochondria provides a useful tool for studying the functional consequences of mitochondrial localization of a protein of interest.
