Indirect Response Mechanism

An indirect response mechanism describes a biological or clinical effect that occurs when an intervention changes an intermediate process, which then alters the measured outcome rather than acting on it directly. In pharmacology, a drug may inhibit or stimulate the production or loss of a response mediator, such as a biomarker, cell population, or physiological signal; the response therefore develops over time according to mediator turnover and may persist after drug concentrations decline. Indirect response models help characterize delayed onset, hysteresis, accumulation, and recovery in clinical studies. They support dose selection, exposure-response analysis, treatment scheduling, and interpretation of biomarkers when drug effects do not track plasma concentrations immediately.
