Clonal Hematopoietic Analysis

Clonal hematopoietic analysis is the study of how individual blood-forming stem and progenitor cells generate descendant cell populations, revealing patterns of lineage development and blood production. Researchers identify and track clones using genetic markers, naturally occurring mutations, or experimental barcodes, then compare marker patterns across hematopoietic cells to reconstruct lineage relationships and assess clonal expansion. In developmental biology, this approach clarifies how stem cells balance self-renewal with differentiation into diverse blood lineages and how those decisions change over time. It also supports investigations of tissue regeneration, hematopoietic disorders, and the cellular origins of abnormal blood populations.

This method allows non-invasive fate mapping of spectrally-coded HSPCs-derived cells in intact tissues for extensive periods of time following transplantation.

We have identified the placenta as a major hematopoietic organ during development. We found that hematopoietic stem cells (HSCs) are both generated and expanded in the placenta in unique microenvironmental niches. Here, we describe experimental techniques required for isolation and visualization of HSCs in the mouse placenta.

Cell organization of craniofacial bones has long been hypothesized but never directly visualized. Multi-spectral cell labeling and in vivo live imaging allows visualization of dynamic cell behavior in zebrafish lower jaw. Here, we detail the protocol to manipulate Zebrabow transgenic fish and directly observe cell intercalation and morphological changes of chondrocytes in the Meckel’s cartilage.

Here we present methodology for the clonal analysis of hematopoietic stem cell precursors during murine embryonic development. We combine index sorting of single cells from the embryonic aorta-gonad-mesonephros region with endothelial cell co-culture and transplantation to characterize the phenotypic properties and engraftment potential of single hematopoietic precursors.

A method to analyse the distribution of bone marrow hematopoietic progenitors in flow cytometry as well as to efficiently isolate highly purified hematopoietic stem cells (HSCs) is described. The isolation procedure is essentially based on magnetic enrichment of c-Kit+ cells and cell sorting to purify HSCs for cellular and molecular studies.
