Cryopreservation Storage

Cryopreservation storage is the long-term preservation of cells, tissues, embryos, gametes, or other biological materials at ultra-low temperatures, allowing developmental biology samples to remain available after collection. Before storage, cryoprotective agents help limit cellular injury, while controlled-rate cooling or vitrification reduces ice-crystal formation; samples are then maintained in liquid-nitrogen conditions where biochemical activity is greatly slowed. This approach supports the banking of embryos, germ cells, stem cells, and developmental tissues for later analysis, transplantation, or genetic studies. Reliable storage preserves scarce specimens, enables repeatable experiments across time, and helps researchers compare developmental states without continuously collecting new material.

Here we present a low cost, durable cryotolerant device for sample retrieval from Dewar tanks filled with liquid nitrogen. The ease of construction and modular design of the device makes the process of sample retrieval from cryogenic tanks safe and easy.

In this video, we demonstrate an ultra-rapid cryo vitrification method for preserving biological samples. The method entails the use of a combination of cryoprotectants to protect the biological tissue from freezing damage by preventing intracellular ice crystal formation.

Mouse Sperm Cryopreservation and Recovery using the I·Cryo Kit

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

Isolation and Cryopreservation of Neonatal Rat Cardiomyocytes

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.

Cryopreservation of Preimplantation Embryos of Cattle, Sheep, and Goats

Preimplantation embryos may be cryopreserved after placement into a hypertonic cryoprotective solution to cause cellular dehydration. After equilibration, ice crystal formation is induced in the solution surrounding the embryo. Further dehydration occurs as the embryo is slowly cooled to subzero temperatures before plunging into liquid nitrogen for storage.
