Multichannel Imaging

Multichannel imaging is a microscopy approach that captures signals from multiple fluorescent labels or optical modalities in the same specimen, allowing distinct structures, molecules, or cell behaviors to be examined together. It works by exciting or detecting each channel under defined spectral or optical conditions and separating the resulting signals through filters, detectors, or computational unmixing; registration aligns channels for comparison across space and time. In developmental biology, this approach reveals how cell populations interact, migrate, divide, and specialize during tissue formation. Combined with live imaging and quantitative analysis, it can connect gene expression or signaling activity with morphogenesis and improve interpretation of complex developmental dynamics.

The protocol describes the methodology of extracellular recording in the motor cortex (MC) to reveal extracellular electrophysiological properties in freely moving conscious mice, as well as the data analysis of local field potentials (LFPs) and spikes, which is useful for evaluating the network neural activity underlying behaviors of interest.

Many microfluidic devices have been developed for use in the study of electrotaxis. Yet, none of these chips allows the efficient study of the simultaneous chemical and electric-field (EF) effects on cells. We developed a polymethylmethacrylate-based device that offers better-controlled coexisting EF and chemical stimulation for use in electrotaxis research.

We detail a new near-infrared fluorescence (NIRF) catheter for 2-dimensional intravascular molecular imaging of plaque biology in vivo. Hybrid µCT-FMT imaging and image analysis

We describe a protocol for hybrid imaging, combining fluorescence-mediated tomography (FMT) with micro computed tomography (µCT). After fusion and reconstruction, we perform interactive organ segmentation to extract quantitative measurements of the fluorescence distribution.
