Defined Culture Conditions

Defined culture conditions are laboratory environments in which the chemical, physical, and nutritional inputs supporting cells or tissues are specified and controlled, making developmental responses more reproducible. In developmental biology, researchers regulate components such as basal medium, growth factors, temperature, pH, oxygen, substrate, and cell density; changing these variables can alter signaling pathways, proliferation, differentiation, and tissue organization. This control helps separate the effects of individual cues during embryonic, stem-cell, and organoid studies, enabling comparisons across experiments and refinement of models for development, disease, and regenerative research.

The production of specialized retinal cells from pluripotent stem cells is a turning point in the development of stem cell-based therapy for retinal diseases. In this protocol, we describe a stable, highly efficient differentiation strategy for the generation of postganglionic sympathetic neurons from human pluripotent stem cells. This model will make neurons available for the use of studies of multiple autonomic disorders.
