Ex Vivo Tissue

Ex vivo tissue refers to tissue removed from an organism and maintained outside the body under controlled laboratory conditions. By supplying appropriate nutrients, oxygen, temperature, and signaling conditions, researchers preserve tissue viability and, in some cases, its native cellular organization and function. In developmental biology, ex vivo tissue systems allow direct observation of growth, differentiation, morphogenesis, and cell-to-cell interactions while reducing the complexity of the whole organism. These models support studies of developmental mechanisms, tissue responses, and gene function, and can complement in vivo experiments by enabling precise imaging, manipulation, and experimental control.

This is a guideline for constructing in vivo vascularized tissue using a microsurgical arteriovenous loop or a flow-through pedicle configuration inside a tissue engineering chamber. The vascularized tissues generated can be employed for organ regeneration and replacement of tissue defects, as well as for drug testing and disease modeling.

Oncolytic viruses are promising for cancer therapeutics. The ability to ascertain the infectability of live tissue specimens obtained from patients prior to treatment is a unique advantage of this therapeutic approach. This protocol describes how to process tissues for ex vivo infection with oncolytic virus and subsequent viral quantification.
