Islet Coordination

Islet coordination is the collective regulation of endocrine cell activity within pancreatic islets, enabling these cell clusters to respond coherently to changing nutrient levels and maintain glucose homeostasis. Coordination arises through communication among insulin-, glucagon-, and somatostatin-producing cells, supported by shared blood supply, paracrine signals, and electrical coupling that synchronize hormone secretion. In developmental biology, studying islet coordination reveals how cellular organization and intercellular communication emerge as the endocrine pancreas forms. These insights support research into pancreatic development, diabetes, stem cell-derived islets, and strategies for improving the function of engineered or transplanted insulin-producing tissue.

Pancreatic islets, also called the Islets of Langerhans, are a cluster of endocrine cells which produces hormones for glucose regulation and other important biological functions. This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.

Effective Isolation of Functional Islets from Neonatal Mouse Pancreas

We describe a protocol herein for isolating intact islets from neonatal mice. Pancreata were partially digested with collagenase, followed by washing and hand picking. 20 - 80 islet clusters can be obtained per pancreas from newly born mice, which are suitable for several islet studies.
