Stereology Technique

Stereology is a quantitative technique that estimates three-dimensional features, such as cell number, volume, length, and surface area, from two-dimensional tissue sections, making complex biological structures measurable and comparable. It works by combining systematic random sampling with geometric probes, including point counting and the optical disector, to reduce bias when specimens are sectioned and analyzed microscopically. In developmental biology, stereology can quantify changes in organ growth, cell proliferation, neuronal populations, and tissue organization across developmental stages. These measurements strengthen links between developmental mechanisms and structural outcomes while supporting rigorous comparisons among experimental conditions.

Here, we present a stereological method, the optical fractionator, used to quantify the formation of new neurons, and their survival, in the rat hippocampus following electroconvulsive stimulation. When correctly implemented, the sensitivity and efficiency of stereological methods ensures accurate estimates with a fixed and predetermined precision.

Brain myeloid cells characterization following stroke can be performed by stereology using the optical fractionator method, or by a flow cytometric analysis of brain leukocytes suspensions. Both are useful techniques to perform an accurate phenotypical distinction of the main myeloid cell subsets found in the ischemic brain.
