Metaphase Ii Stage

The Metaphase II stage is a phase of meiosis II in which duplicated chromosomes align at the cell’s equatorial plane before sister chromatids separate. In mammalian oocytes, spindle fibers organize and attach to chromosome kinetochores, while the secondary oocyte typically remains arrested at Metaphase II until fertilization triggers cell-cycle resumption, chromatid separation, and extrusion of the second polar body. Understanding this stage is important in developmental biology because it links oocyte maturation with fertilization and early embryonic development. Its assessment also supports research on gamete quality, chromosome segregation, aneuploidy, and assisted reproductive technologies.

DNA-FISH Metaphase Spreads to Distinguish Extrachromosomal DNA from Homogeneously Staining Regions in Human Cancer Cell Lines

We report a DNA-FISH metaphase spread protocol that visually detects locus copy number and location within the genome. This approach enables single-cell resolution of amplification states, specifically in cancer cell lines containing extrachromosomal DNA and homogeneously staining regions.

Simultaneous extracellular long term recordings from two different brain neuropiles or two different anatomical tracts were established in honeybees. These recordings allow the investigation of temporal aspects of neuronal processing across different brain areas at the single neuron as well as at the ensemble level in a behaving animal.

This manuscript describes chromatin spread preparation methods for mouse prophase, metaphase I and II-staged oocytes. These fundamental techniques allow for the study of chromatin-bound proteins and chromosome morphology throughout mammalian oogenesis.
