Ovarian Cortex Preparation

Ovarian cortex preparation is a tissue-processing method that isolates the outer region of the ovary, where ovarian follicles and supporting cells are organized, for developmental and reproductive research. The procedure typically involves carefully dissecting the cortex from medullary tissue, trimming it into defined fragments, and preserving its architecture through appropriate handling or culture conditions. Prepared ovarian cortex enables researchers to examine follicle activation, growth, and maturation while studying interactions between germ cells and the surrounding somatic environment. In developmental biology, this approach supports investigations of ovarian development, fertility preservation, reproductive aging, and the effects of genetic or environmental factors on follicle development.

In vitro culture of bovine ovarian cortex and the effect of nutritional Stair-step diet on ovarian microenvironment is presented. Ovarian cortex pieces were cultured for seven days and steroids, cytokines, and follicle stages were evaluated. The Stair-Step diet treatment had increased steroidogenesis resulting in follicle progression in culture.
