Sers Substrates

SERS substrates are engineered surfaces that amplify Raman scattering from molecules adsorbed on or near a nanostructured material, enabling highly sensitive chemical detection. Typically made from gold or silver nanostructures, they concentrate incident light through localized surface plasmon resonances, while close contact between the analyte and metal surface can provide additional chemical enhancement. In engineering, substrate geometry, composition, spacing, and surface chemistry are designed to improve signal intensity, reproducibility, and selectivity. These platforms support trace analysis, materials characterization, environmental monitoring, biomedical sensing, and the development of compact analytical devices.

This video demonstrates a quantification technique for CO2 released during substrate oxidation using 14C-radiolabeled substrates. The released 14CO2 is trapped in an alkaline solution and quantified by a scintillation counter. The oxidation of different substrates varies between tissues and reflects the pathophysiological condition of the tissue.

CRISPR/Cas systems mediate adaptive immunity in Bacteria and Archaea. Many Cas proteins are proposed to act as endoribonucleases acting on crRNA precursors of varying length. Here we illustrate three different approaches to generate pre-crRNA substrates for the biochemical analysis of Cas endonuclease activity.
